I have constructed a relatively large shuttle vector that can be propagated in both Gram-positive (E. faecium) and Gram-negative bacteria (E. coli). I used to have trouble transforming this plasmid vector into other commercially available E. coli-competent cells. Then, I came across this fantastic product that can accommodate and propagate larger plasmids.
This product is useful in maintaining large vectors.

For consistent transformation efficiency, do not reuse the cells once thawed from -80 degrees C.
Using this kit, I have harnessed a better transformation efficiency for my large shuttle vector. Due to the increased transformation efficiency, I screened more colonies and got more positive clones.
