This is a recombinant Human IFN-Y, which I used for activation of human monocyte cell line. THP-1 monocytes are differentiated into macrophages by 24 h incubation with 150 nM phorbol 12-myristate 13-acetate (PMA, Sigma, P8139) followed by 24 h incubation in RPMI medium. Macrophages were polarized in M1 macrophages by incubation with 20 ng/ml of IFN-Y.

Regarding differentiation and activation of monocytes and macrophages, do not exceed the amount that is recommended by protocol. It causes over activation.

Results Summary

Adding IFNg led to activation of THP-1 cells and I used them for cytotoxicity assay. After differentiation of THP-1 cells, they will attach to the plate.
