I am interested in studying the molecular mechanism of hedgehog signal transduction in vertebrate cells. I routinely perform western blotting for target genes and core hedgehog pathway components to measure the signaling output and monitor baseline levels, respectively. We use the Bio-Rad power supply equipment for running gels and during transferring proteins to nitrocellulose membranes. We had purchased this power supply equipment based on our own positive experience with Bio-Rad products in this category.

Protocol Overview

This power supply unit is used during SDS-PAGE gel running step and also during protein transfer to nitrocellulose membranes. For SDS-PAGE step, load samples in wells equilibrated with 1x running buffer and run at constant voltage 120–140 V. For protein transferring step, I use wet-transfer technique. After preparing the gel-nitrocellulose membrane/Whatman paper sandwich, carefully place the cassette in the wet-transfer box containing 1x transfer buffer and run at constant current of 200 mA (400 mA for 2 gels).
