The overall goal of this research was to understand protein targeting to specific cellular domains. More specifically with this experiment I was studying how different protein modifications influence protein localization in the lipid compartments. This product was chosen due to availability within the lab.

Starting Material

Cell lysates from trypanosoma cruzi cells

Protocol Overview

The cell lysates were made from trypanosoma cruzi cells transfected to express different proteins of choice. Cells were mixes with 2x sample buffer and incubated at 95 degrees C for 10 minutes. The cell lysates were than loaded on the 12% gel from BioRad and were run on Criterion Blotter (represented in this review) on 100 volts for30 minutes or until the dye front reached the bottom of the gel.

Tips

Use higher voltage for first couple of minutes to help lysates get into the well and then run at the lower voltage that is consistent throughout the experiment.

Results Summary

Proteins of various sizes were observed after the gel transfer. There were no artifacts observed and the gel run very straight.
