Description

The Sequel sequencer system uses single molecule, real time (SMRT) sequencing technology that utilizes immobilized DNA polymerase/template complex in tens of thousands of small wells (called zero-mode waveguides). These wells have the ability to detect changes in light emission that arise from processing of nucleotides that are each linked to a different colored fluorophores. As each nucleotide is incorporated by the polymerase through natural DNA synthesis, the fluorescent emission from the fluorophore release are detected in parallel by the wells, generating sequence data. The Sequel system features 1 million zero-mode waveguides per SMRT cell and reports an average read length of over 10,000 bp and a total output of 7.6 Gb. It can process a SMRT cell in as little as 30 minutes to 10 hours and can flexibly run between 1 to 16 cells at a time. Other features include a touchscreen interface, a workdeck, and robotic workflow management.
