In my current work, I express recombinant proteins tagged with HA in mammalian cells to observe condensate formation under osmotic stress. Due to the absence of appropriate antibodies for my protein of interest, the use of a small HA tag allows for visualization via immunofluorescence.

Confocal imaging
The image provides a clear depiction of the successful labeling of the overexpressed protein, highlighted in green fluorescence, against the background of the DAPI-stained nucleus in blue. Notably, the specificity of the antibody is evident from the discernible staining confined solely to the transfected cells. When conducting multicolor imaging, the use of an unconjugated primary antibody proves advantageous, as it offers flexibility in selecting from a diverse array of conjugated secondary antibodies for immunofluorescence staining, thereby optimizing experimental design and ensuring precise visualization of multiple targets within the same sample.
